The abil­i­ty to inhib­it the activ­i­ty of a trypsin unit is called an inhibitor activ­i­ty unit (U).

Trypsin unit

One trypsin unit (micro katal) is defined to hydrolyze 1 μmol of N-ben­­zoyl-L-argi­­nine eth­yl ester (BAEE) per second.

Intro­duc­tion

Soy­bean trypsin inhibitor is a ser­ine pro­tease inhibitor, which can inhib­it the activ­i­ty of trypsin, chy­motrypsin, and plas­min. The abil­i­ty to inhib­it trypsin is high­er than that of chy­motrypsin and plas­min. In addi­tion, it can inhib­it plas­ma kallikrein, clot­ting fac­tor Xa, but can’t inhib­it met­al­lo­pro­teinase, tis­sue kallikreins, acid pro­tease, and sul­­fur-con­­tain­ing pro­tease. Inhibitors and pro­tease can be effec­tive­ly com­bined in equimo­lar amounts to form sta­ble com­plex­es to block the active site of the enzyme. A suit­able pH range for inhibit­ing trypsin is pH 6.0-10.0. Recom­bi­nant soy­bean trypsin inhibitor was expressed by recom­bi­nant E.coli and puri­fied with­out ani­­mal-derived con­t­a­m­i­na­tion and no oth­er protease.

Instruc­tion

Inhibitors and pro­tease can be com­bined in equimo­lar amounts, the rec­om­mend­ed pH for com­bi­na­tion is pH>6.0 , inhibitors and pro­tease won’t be com­bined when pH<5.0.
