Known Appli­ca­tion

Bovine B4GALT1(Y289L) is a mutant galac­to­syl­trans­ferase capa­ble of effi­cient­ly trans­fer­ring GalNAc or Gal­NAz onto ter­mi­nal Glc­NAc residues. This enzyme is par­tic­u­lar­ly use­ful in the syn­the­sis of anti­­body-drug con­ju­gates (ADCs), which are applied in tar­get­ed can­cer ther­a­pies and oth­er bio­med­ical applications.

1.Reac­tion Sys­tem :
50 mM Tris-HCl (pH 7.5), 6 mM MnCl₂

2.Reac­tion Reagents :
① UDP-Gal­­NAz : Anti­body (molar ratio) = 300:1
② Enzyme addi­tion : Enzyme/​Antibody = 5%

3.Prepare the reac­tion mix­ture accord­ing to the con­di­tions above and incu­bate at 37°C for 48 hours. Sub­se­quent­ly, place the reac­tion mix­ture at -15 to -25°C to ter­mi­nate the reac­tion and pro­ceed with sam­ple analysis.
